GLP-1s, longevity and anti-aging claims

A longer life is a hard outcome, and only a few trials have counted deaths. These articles separate those results, all in people who were already ill, from biomarkers and epigenetic clocks, which are surrogates, and from the marketing that treats one as the other.

  1. Are GLP-1s Longevity Drugs? What the Death and Outcome Data Show

GLP-1 drugs lower deaths from any cause by about 12% in people with heart disease, diabetes or kidney disease. No trial has tested them in healthy adults or measured lifespan. SELECT, FLOW, LEADER, dementia, cancer and muscle, in one place.

  2. Does Semaglutide Slow Aging? What the Longevity Evidence Shows

No trial has tested whether semaglutide slows aging. SELECT and FLOW found fewer deaths from any cause in people with heart or kidney disease, and the only aging-clock result is a post hoc 32-week analysis in 84 people with HIV.

  3. Tirzepatide and Longevity: What the Death and Aging Data Show

No study shows tirzepatide extends life. In SURPASS-CVOT, 8.5% died on Mounjaro vs 10.1% on dulaglutide over four years, an exploratory result. Deaths in obesity without diabetes are unreported until 2027, and no aging-clock data exist.

  4. Metformin vs GLP-1 for Longevity: The TAME Trial Has Not Started

No trial shows metformin extends life in people without diabetes. TAME, the metformin aging trial, has no registry record and was still unfunded at last report. Semaglutide has randomized outcome data, but only in people already at high risk.

  5. NAD+ and Longevity: What the NMN and NR Trials Actually Show

NMN and nicotinamide riboside reliably raise blood NAD+, but human trials on muscle, blood sugar, blood pressure and cognition are mostly null, and none measured lifespan. Trial-by-trial effect sizes, IV NAD+, and FDA's 2025 reversal on NMN.
